Is it safe for me to update Plugin while server is running?
Also will the Data file be rest if I delete old Plugin file and upload new plugin file to server?

Sure, just download it and install it as you normally would. Unless you delete the data yourself, it isn't going to be removed.
Would it need a server restart once I've replace the file? I've experience where oxide.reload wouldn't work and it just goes blank when I type /s
No, plugin updates do not require a restart generally. If a reload does not work, then I'd suggest checking your oxide/logs for errors.
